The MSP nanodisc is composed of α-helical-amphipathic proteins derived from apolipoprotein A-I, and phospholipid molecules, in which α-helical-amphipathic proteins wrap around the lipid bilayer as a homo dimer. These amphipathic proteins are commonly called membrane scaffold proteins (MSP), and they act as a belt around the lipid bilayer, holding them together and stabilizing the nanodisc. And their length determines the diameter of the nanodisc. As mentioned above, MSP is amphiphilic, and its structure contains a hydrophobic end towards the inner phospholipid layer and a hydrophilic end towards the outer layer. Such structure enables MSP nanodisc to have high solubility in aqueous solutions. Once the membrane protein is integrated into the MSP nanodisc, the membrane protein can maintain its monodisperse and activity in solution without detergent, which provides a strong technical support for membrane protein research. It can be seen that membrane scaffold protein (MSP) is the key component of nanodisc.

MSP1D1 is one of the most popular constructs, and is created by removing the first 11 N-terminal amino acids from MSP1 and replacing the 6×His tag and X linker with a 7×His tag and tobacco etch virus protease (TEV) linker to allow affinity tag removal. MSP1D1 can create assembled nanodiscs 9.5 nm in diameter. In addition, MSP1D1 can be modified further to create MSP1E1D1, MSP1E2D1, and MSP1E3D1 to enable nanodiscs with increased sizes (10.5 nm, 11.1 nm, and 12.1 nm in diameter, respectively). MSP2N2 and MSP2N3 represent other two popular constructs that can create assembled nanodiscs 16–17 nm in diameter. Smaller MSPs, MSP1D1ΔH4, MSP1D1ΔH5, MSP1D1ΔH4–5, and MSP1D1ΔH4–6, have also been designed to create nanodiscs with decreased sizes [1].
